Why test for Campylobacter?
Campylobacter is recognized by food safety regulators as one of the most important contaminants of food and is the leading bacterial cause of human gastroenteritis. Campylobacter infections can be fatal. Low infective dose: as few as 500. Campylobacter cells can cause sickness.

Poultry and raw milk are the main source of Campylobacter. Thus, dairy products, poultry and ready-to-eat meals have been the main causes of outbreaks in recent times. Water contaminated with Campylobacter has also led to Campylobacteriosis.

Processed poultry products can also be prone to post-processing contamination. Campylobacter will not grow below 30°c. However, they will survive better under chilled conditions than at ambient temperatures, and can multiply once the food is removed from refrigeration. Accordingly, it is very important to ensure that chilled foods are free of Campylobacter.

Captivate™ Salmonella

Captive™ is a range of antibody coated paramagnetic particles for the specific ImmunoMagneticSeparation of microorganisms.

This technology consists of microscopic paramagnetic particles. The beads have a magnetite core and a “ceramic” zirconium oxide coating. The beads are manufactured by high speed blending process and typically cover a size diameter range of 1-4 µm, with a 2.5 µm average size.

Purified antibodies to surface components of the target microorganism are covalently coupled to the bead. With careful antibody selection, a highly specific separation system for microorganisms is produced.

The pre-coated beads are designed for the IMS of target bacteria from enrichment cultures. A sample is taken form a filter stomacher bag and incubated with the Captive™ beads for 30 minutes. The bead/microoragnism complexes are then removed from the sample by placing the sample in a magnetic concentrator device. This separates them form the background organisms and interfering materials. The complexes are then washed using a PBS/Teen 20 wash buffer to remove non-specifically bound material. The beads can then be plated out onto the appropriate selective agar media and incubated as described.

The IMS technique will increase the sensitivity of the methodology and, in most circumstances, results can be achieved 24 hours earlier than standard protocols.

These products can also serve as a capture system for rapid detection systems.

Special Notes on IMS Techniques.
There are important factores that affect the performance of IMS techniques. Thorough mixing of the particles and sample allied with efficient recovery of the beads from the sample matrix is paramount to the success of this technique. Care must be taken not to aspirate the sample vigorously as this can result in the loss of captured target organisms. Certain sample types (e.g. very fatty, particulate and viscous samples) can interfere with bead recovery. To counter-act this interference, samples can also be diluted in PBS-Tween e.g. 1:2-1:4, reducing the effect of the matrix and allowing more efficient bead recovery. Alternatively with problem samples, after the initial magnetic separation the incomplete removal of the sample (i.e. remove 800 µl) and continuation of the wash protocol as described can minimise bead losses.

Captive™O157 are magnetisable particles coated with specific antibody intended for the isolation of E.coli O157:H7 from food, animal feeds, beverages, pharmaceutical or environmental samples. The particles help to concentrate O157:H7 cells in mixed culture reducing the probability of missing low numbers or overgrowth of O157:H7 colonies by competing flora. In fact, immunomagnetic separation is now regarded as the gold standard method for isolation of E.coli O157:H7 from food and environmental samples.

E.coli O157:H7 is the primary serovar associated with food borne gastrointestinal infection, resulting in self-limiting diarrhoea, that can lead to serious disease conditions such as haemorrhagic colitis, haemolytic uraemic syndrome and thrombotic thrombcocytopaenic purpura (TTP).

The organism itself is associated with raw meats and unpasteurised milk, probably due to the implication of farm animals and particularly cattle as carriers of E.coli O157:H7. Large outbreaks have been recorded in the United States from consumption of unpasteurised apple juice (apple cider) possibly as a result of using apples which have fallen to the ground where the potential for contamination with the organism exists.

Why test?
Staphylococcal food poisoning is one of the most common types of foodborne disease worldwide. It is also one of the most economically important diseases, estimated to cost the United States approximately $1.5 billion each year.

Some strains of S.aureus are capable of producing a highly heat-stable protein toxin that causes illness in humans. Heat treatments commonly used in food processing may kill viable S.aureus cells but cannot be relied upon to inactivate enterotoxins. Therefore, it is advisable that foods be examined for the presence of residual toxin in addition to viable organisms.

A rapid and simple screening test for indicating the presence of Bacillus Diarrhoeal Enterotoxin (BDE) in food and food-related samples, and enrichment cultures.

Why are BDE-Producing Bacillus spp. considered a significant problem to the food industry?

Bacillus cereus and other Bacillus spp. Are frequently found in a very wide range of foods, including baby foods, cooked and frozen rice products, cereals, meat products, soups, milk and milk powders, and extended shelf-life cooked-chill meals.
Food poisoning outbreaks involving B. cereus have resulted in major product recalls and substantial economic loss. In the USA published data indicates that B. cereus causes up to 84,000 cases of food poisoning each year.
The heat resistant nature of Bacillus spores allows them to survive in many foods, which have undergone moderate heat processing and normal cooking processes.

Pathogen & Spoilage Organisms
Contamination of cosmetics, toiletries and pharmaceuticals by Pseudomonas spp. is well documented. Pseudomonas spp. have been linked to consumer infection and product spoilage, resulting in substantial economic loss.

In the European Union, P.aeruginosa must not be detectable in 0.5 g or 0.5 ml of cosmetic products for the eye area, mucous membranes or for cosmetic products specifically intended for children under 3 years. In other cosmetics, P.aeruginosa must not be detectable in 0.1 g or 0.1 ml (EC 1999).

Pseudomonas spp. are associated with the spoilage of a range of food types including dairy products, eggs, fish, meat and vegetables.

Water has been associated with outbreaks or sporadic cases of foodborne illness resulting from Pseudomonas spp. 2 Studies have shown Pseudomonas spp. to be present in ground and spring water, municipal treated water, filters and softeners and bottled water.

The Australasian Bottled Water Association (ABWI) has set standards (ABWI Model Code) stating that P.aeruginosa shall not be detected in five 250 ml samples.

Coagulase test
The enzyme coagulase, produced by a few of the Staphylococcus species, is a key feature of pathogenic Staph. The enzyme produces coagulation of blood, allowing the organism to "wall " its infection off from the host's protective mechanisms rather effectively.

There are 2 methods: slide test and tube test. The tube test is more accurate, but the slide test is faster. If you have a Staph unknown, you might consider running the slide test, and, if negative, running the tube test. The tube method is THE definitive test.

Objectives
Identify the presence of the enzyme coagulase.

Materials needed
0.5 ml rabbit plasma per unknown

The procedure
Pathogenic Staph species (e.g. Staph aureus) can be confirmed using the coagulase test. Since there are 2 kinds of coagulase enzyme—bound and free---there are 2 different tests that can be used to identify these enzymes. Both of the enzymes activate fibrinogen in plasma, in different ways.

Slide Agglutination test
Make a 2,54 cm diameter circle on a clean glass slide using a wax pencil. Place two drops of thawed rabbit plasma into the circle, using a wooden pick or a clean loop. Add a single colony and emulsify it in the plasma. Add a single colony and emulsify it in the plasma. Fibrin threads form between the cells, causing them to agglutinate, or clump. There will a visible clumping of cells within 10-15 seconds. This test is for the bound coagulase enzyme.
Tube Agglutination test
Inoculate a tube with a 0,5 ml of rabbit plasma with the bacterial inoculum. Place at 37 °C and check at 0,5 hour and after (some strains will give a + reaction in a few hours, other strains take longer) by tipping the slide at an angle. Any degree of coagulation is considered a positive test for the free coagulase enzyme. Add a single colony and emulsify it in the plasma. Fibrin threads form between the cells, causing them to agglutinate, or clump. There will a visible clumping of cells within 10-15 seconds. This test is for the bound coagulase enzyme.


Staphylococcus Aureus Visual Immoassay
A rapid and specific screening test for the detection of Staphylococcus aureus in food, cosmetic and pharmaceutical products, and environmental samples.

Staphylococcal food poisoning is one of the most common types of foodborne disease worldwide. Food poisoning outbreaks caused by S. aureus have involved a wide range of foods including meat, poultry and egg products, cream-filled bakery products and dairy products.

Because of the pathogenicity of S. aureus, the United States Pharmacopeia (USP) requires its absence in certain nonsterile pharmaceutical raw materials and finished products (U.S. Pharmacopeia 1995).

In the European Union, S. aureus must not be detectable in 0.5 g or ml of cosmetic products specifically intended for: children under 3 years, eye area and mucous membranes. In other cosmetics, S. aureus must not be detectable in 0.1 g or ml (EC 1999).

Some strains of S. aureus produce enterotoxins, which show a high degree of thermal stability when subjected to heat treatments commonly used in food processing to kill viable cells. Therefore, it is advisable that foods be examined for the presence of residual toxin using a Staphylococcal Enterotoxin detection kit.

The same enrichment broth, used to detect S.aureus, can also be tested for Staphylococcal Enterotoxins using the Staphylococcal Enterotoxin VIA or UNIQUE™ Staphylococcal Enterotoxin assay.
Individual toxins can then be identified using the Staphylococcal Enterotoxin Identification VI
